Sensitive skin is a condition of hyperreactivity to environmental factors or topically applied products. It is characterized by heightened sensitivity and reactivity, often resulting in discomfort, irritation, redness, itching, stinging, burning, and may be accompanied by erythema. Soothing cosmetics are aimed to alleviate symptoms such as burning, stinging, itching, tightness, erythema, scaling, and redness (capillary dilation) that result from skin barrier damage, abnormal nerve function, and inflammatory reactions.

  • Neutrophil count

Skin inflammation is the main skin irritation phenotype of cosmetics. Exposure to Sodium Laurylsulfonate (SLS) may elicit an irritant response in organisms. Irritant enters zebrafish to induce an inflammatory response, and neutrophils undergo an immune response, migrating and aggregating to the skin epidermis. At Creative Biogene, changes in the number of neutrophils in the skin epidermis of a transgenic neutrophil green fluorescent strain of zebrafish (MPX) are used to evaluate skin soothing efficacy of cosmetics.

  • Inflammatory factor expression

TNF-α is produced in large quantities when the body is stimulated, mainly from activated macrophages, natural killer cells, and T lymphocytes. IL-6 can promote the upregulation of neutrophil function, and promote and regulate the secretion of inflammatory mediators such as cytokines, adhesion molecules, and nitric oxide. IL-8 is secreted and produced by monocytes and vascular endothelial cells, with levels rapidly increasing during inflammation. At Creative Biogene, we evaluate the skin soothing efficacy by measuring the mRNA level of inflammatory factors. A reduction in the mRNA level of inflammatory factors in response to your cosmetics indicates the soothing efficacy of your cosmetics.

  • Tryptase content

Mast cells are the main effector cells in allergic reactions, and the number of activated mast cells is correlated with the severity of allergies. Tryptase is an enzyme found mainly in mast cells. After degranulation of mast cells, Tryptase is converted into an active conformation and released into the extracellular space. Tryptase can act as a marker for mast cell degranulation.
