Overview

Our Conditional & Inducible Systems Generation service supports the development of custom zebrafish lines for tissue-specific gene expression, time-controlled genetic manipulation, permanent cell labeling, and lineage-tracing studies.

Using established Tol2 transgenesis, Gal4/UAS, heat-shock induction, and Cre-based recombination platforms, we help researchers select and combine appropriate genetic tools according to their target tissue, experimental timing, desired readout, and downstream application.

These systems are suitable for developmental biology, regeneration, neuroscience, disease modeling, cell biology, and in vivo functional studies.

Why Use Conditional and Inducible Zebrafish Systems?

Constitutive whole-body expression of a transgene may cause early developmental phenotypes, toxicity, or effects that obscure the role of a gene in a defined tissue or developmental stage. Conditional and inducible systems provide additional experimental control over:

Where a gene, reporter, or effector is active;

When an expression or recombination event is initiated;

Whether the outcome is transient or permanent;

Which cell populations are labeled and followed over time;

How a single tissue-specific driver can be combined with multiple downstream effectors.

Core Service Platforms

Tol2-Mediated Tissue-Specific Transgenic Line Generation

Tol2 transposon-mediated transgenesis is a widely used approach for generating stable zebrafish transgenic lines. A tissue- or cell type-specific promoter is used to drive a reporter gene, gene of interest, recombinase, transcriptional activator, or other functional effector.

Key Features

  • Direct and well-established route for tissue-specific transgene expression;
  • Compatible with a broad range of tissue-specific promoters and effectors;
  • Suitable for building stand-alone expression lines or driver lines for binary systems;
  • Supports stable germline-transmitted transgenic line generation.

Gal4/UAS Binary Expression System

The Gal4/UAS system separates tissue specificity from downstream gene function. A Driver line expresses Gal4 in the target tissue, while a separate Responder line carries a UAS-regulated reporter or effector gene. Crossing the two lines activates UAS-dependent expression in the Gal4-positive cell population.

Key Features

  • A single driver line can be crossed with multiple UAS responder lines;
  • Enables flexible replacement of reporters or effectors without rebuilding the tissue-specific driver;
  • Suitable for laboratories seeking a modular and expandable genetic toolkit;
  • Can be combined with inducible components, such as heat-shock-regulated Gal4 expression or other custom strategies.

Heat-Shock Inducible hsp70l System

The zebrafish hsp70l promoter is commonly used to initiate transgene expression in response to controlled heat-shock treatment. This system is particularly useful when transient expression needs to be initiated at a defined developmental or experimental time point.

Key Features

  • Provides a practical approach for initiating expression at selected time points;
  • Particularly suitable for embryonic and larval studies;
  • Can be used as a transient expression system or as an upstream trigger for permanent Cre/loxP recombination;
  • Does not require administration of a chemical inducer.

Cre/loxP Reporter, Conditional Activation, and Lineage-Tracing Systems

Cre/loxP recombination enables irreversible DNA recombination between loxP sites. In zebrafish, this system is commonly used to activate a reporter or effector after Cre-mediated removal of a loxP-flanked STOP cassette.

This approach is particularly useful when a transient Cre signal needs to be converted into permanent reporter expression or long-term genetic activation.

Key Features

  • Converts Cre activity into a durable genetic outcome;
  • Supports permanent labeling of targeted cells and their progeny;
  • Compatible with tissue-specific Cre or inducible CreERT2 drivers;
  • Can be integrated into multi-line genetic strategies.

CreERT2 + 4-OHT Inducible Recombination System

CreERT2 combines Cre recombinase with a modified estrogen receptor ligand-binding domain. In the absence of inducer, CreERT2 activity is generally limited. Following treatment with 4-hydroxytamoxifen (4-OHT), CreERT2 can enter the nucleus and mediate loxP recombination.

When paired with a loxP responder line, this system provides tissue-specific and time-controlled permanent genetic activation.

Key Features

  • Combines tissue specificity with chemical induction;
  • Enables researchers to select the time window for recombination;
  • Produces a permanent recombination outcome after successful induction;
  • Well suited for developmental, adult, regenerative, and cell-fate studies.

Service Workflow

Step 1

Project Assessment & Strategy Selection

Define the target tissue or cell type, intended genetic outcome, and required spatial or temporal control. We then recommend an appropriate platform, such as Tol2 transgenesis, Gal4/UAS, hsp70l, Cre/loxP, or CreERT2.

Step 2

Construct Design

Design the transgenic cassette, including the promoter, reporter or effector, and required regulatory elements. A project-specific screening and validation strategy is established before injection.

Step 3

Transgenic Line Generation

Introduce validated constructs into one-cell-stage embryos by microinjection. Candidate founders are raised and screened using fluorescence, genotyping, or other relevant methods.

Step 4

Germline Transmission Validation

Outcross positive founders and screen F1 progeny to confirm stable germline transmission and identify suitable lines.

Step 5

Optional Functional Validation

Assess tissue-specific expression, driver–responder activity, heat-shock induction, Cre/loxP recombination, or 4-OHT-induced CreERT2 activity, depending on the system design.

Deliverables

Depending on the project design, deliverables may include:

  • Positive founder-derived fish or validated F1 transgenic progeny;
  • Transgene construct map and sequence summary;
  • Genotyping primers and recommended PCR conditions;
  • Germline transmission confirmation;
  • Reporter-expression or molecular screening results;
  • Basic breeding and line-maintenance recommendations.
